Reverse transcriptase acts as a primer of RNA-directed DNA synthesis by binding to the bulge of ε. Encapsidation and replication of HBV may also be impaired by mutations in the upper stem and loop of ε. Mutations at the precore codon 17 (G1899A) have been found alone [35],[36] or together with other precore mutations like G1862T transversion [15] or the 28-stop codon mutation could also lead to HBeAg seronegativity.
